摘 要:目的 研究重组人白细胞介素 1 0 (rhIL 1 0 )对银屑病患者与正常人外周血单个核细胞 (PBMC)产生细胞因子与一氧化氮 (NO)的影响 ,探讨rhIL 1 0治疗银屑病机制。方法 分离、培养银屑病患者与正常人PBMC ,并检测PHA诱导PBMC产生IL 2、IFN γ与LPS诱导PBMC产生IL 1、IL 6、IL 8和NO水平。结果 银屑病患者活化的PBMC产生IFN γ、IL 1、IL 6和IL 8的水平明显高于正常健康人 ,IL 2与NO水平也较正常人为高 ,提示银屑病活动期一些炎性细胞因子水平升高。rhIL 1 0 (2 5、5、1 0、2 0、40 μg/L)对PHA诱导的银屑病患者与正常人PBMC产生IL 2、IFN γ均有抑制作用 ,对LPS诱导银屑病患者与正常人PBMC产生IL 1、IL 6、IL 8和NO同样具有抑制作用 ,并呈明显剂量依赖关系。anti IL 1 0McAb(2 5、5、1 0、2 0、40mg/L)在rhIL 1 0存在与不存在情况下均能促进PHA活化的银屑病患者与正常健康人PBMC产生IL 2及IFN γ ,但银屑病患者PBMC产生上述细胞因子能力低于正常人。结论 rhIL 1 0对银屑病患者PBMC分泌的炎性细胞因子水平升高具有抑制作用 。Objective To observe the effects of recombinant human i nterleukin 10 (rhIL 10 ) on peripheral blood mononuclear cells (PBMC) of psoriasis patients and normal persons. Methods PHA induced PBMC production of IL 2 and IFN r, LPS induced production of IL 1 , IL 6, IL 8 and nitric oxide (NO) were determined. Results It was revealed that the concentrations of IFN γ, IL 1, IL 6 and IL 8 produced by activated PBMC of psoriasis patients were significantly higher than that of normal persons in the absence of rhIL 10. Similarly, IL 2 and NO level were also higher in the former group than the latter one. These data suggest that the selected patients were in active stage of psoriasis. Increased concentrations of rhIL 10 significantly inhibited production of cytokines (IL 2 and IFN γ, IL 1, IL 6, IL 8 and NO) by PBMC in the presence of PHA or LPS in a dose dependent way. There was not any difference ( P> 0 05) between the two groups, indicating their sensitivities to rhIL 10 were similar with each other. Whether rhIL 10 existed or not, anti IL 10MAB (2 5, 5, 10, 20, 40 mg/L) restored production of IL 2 and IFN γ by PHA induced PBMC of both groups. However, the recovery of cytokines production of psoriasis PBMC was less than that of normal people. These results indicated that anti IL 10MAB was able to recognize rhIL 10, as well as natural IL 10 (a negative factor) and the productivity of IL 10 in patient PBMC was impaired. Conclusion RhIL 10 can significantly inhibit the pro inflammatory cytokine production (IL 1, IL 6, IL 8 and NO) as well as the immunological cytokine production (IL 2 and IFNr), suggesting that rhIL 10 has combined immunosuppressive and anti inflammatory activity.
