摘  要:目的 :研究胰岛样生长因子 -Ⅰ受体 (IGF -ⅠR)两亚基 (α ,β)和磷酸化酪氨酸蛋白 (P -Tyr)在增生性瘢痕和正常皮肤中的表达特征。方法 :用免疫组化技术检测这三种蛋白在 1 0例增生性瘢痕和 1 0例正常皮肤中的表达变化规律。结果 :在正常皮肤中 ,IGF -ⅠRα ,IGF -ⅠRβ和P -Tyr的阳性率分别为 (2 9.4± 5 .6) %、(2 4 .2± 4 .6) %和 (46 .8± 7.2 ) %。在增生性瘢痕中 ,三种蛋白的阳性细胞率明显升高 (P <0 .0 1 ) ,其中IGF -ⅠR主要定位于表皮细胞和成纤维细胞上 ,含有P -Tyr蛋白的阳性细胞主要为表皮细胞、内皮细胞和成纤维细胞。结论 :增生性瘢痕形成可能与IGF -R蛋白高表达有关。Objective:To explore the effect of two subunits (IGF-ⅠRα and IGF-ⅠRβ) of insulin-like growth factor-Ⅰ receptor and phosphorylated tyrosine proteins (P-Tyr) on the formation of hypertrophic scars by investigating the localization and expression of these proteins in normal skins and hypertrophic scars.Methods:The expression intensity and distribution of IGF-ⅠRα, IGF-ⅠRβ and P-Tyr were examined with immunohistochemistry and pathological methods in 10 hypertrophic scars and 10 specimens of normal skins.Results:In normal skins, the positive cellular rates of IGF-ⅠRα, IGF-ⅠRβ and P-Tyr were (29.4±5.6)%, (24.2±4.6)% and (46.8±7.2)%, respectively. In hypertrophic scars, the positive cellular rates of these proteins were significantly increased compared with normal skins ( P <0.01). The protein particles of IGF-ⅠR were principally distributed in cellular membrane of epidermal cells and fibroblasts. Positive signals of P-Tyr were chiefly located in epidermal cells, endothelial cells and some fibroblasts.Conclusion:The increment of IGF-ⅠRα and IGF-ⅠRβ contents in hypertrophic scars over normal skins might be one of the mechanisms underlying scar formation.
