摘  要:目的 :研究内皮型一氧化氮合酶 (eNOS)与兔实验性血管痉挛的关系。方法 :2 8只新西兰白兔分成假手术对照组 (8只 )、手术加普通饮食组 (8只 )和手术加高胆固醇饮食组 (12只 )。血管造影观察球囊内皮剥脱前后和分别给予普通饮食和高胆固醇饮食喂养 8周后麦角新碱诱发血管痉挛情况。硝酸还原酶法测定各组血浆一氧化氮 (NO)水平 ,免疫组化检测各组血管eNOS含量。结果 :血管造影显示手术加高胆固醇饮食组可诱导出局限性血管痉挛。免疫组化显示eNOS定位在血管内皮细胞胞质、痉挛血管节段内皮细胞内表达减少。手术加高胆固醇饮食组血浆NO的含量也明显下降。结论 :高胆固醇和内皮剥脱使eNOS合成下调 。Objective:To investigate the relationship between eNOS and experimental vasospasm.Methods:Twenty eight New Zealand white rabbits were randomly allocated into three groups: Sham operation control group (Group1, n=8), balloon endothelial denudation + normal diet group (Group2, n=8) and balloon endothelial denudation + hypercholesterol diet group (Group3, n=12). Angiography was performed to detect the vasospasm induced by ergonovine before and after denudation and 8 week later. Immunohistochemistry were performed to localize and quantitate the expression of eNOS. Serum NO level was detected in 0 week and 8 week later by the way of nitrate reductase.Results:Visible Vasospasm was induced at the denuded sites in group3. And eNOS expression in the same locations was decreased compared with other sites in the other group. Serum NO level in Group3 was also decreased compared with the other two groups.Conclusion:The decrement of eNOS expression resulted from balloon endothelial denudation and hypercholesterolemia may play the important role in the pathogenesis of experimental vasospasm.
