摘  要:目的 研究地氟醚预处理对缺氧/复氧所致的心肌细胞M受体密度变化的影响。方法原代培养SD幼鼠的心肌细胞,至第3天时,随机分为五组,组1正常对照(n=6):细胞不经任何处理;组2短期缺氧/复氧(n=6):细胞缺氧2h,复氧1h;组3地氟醚预处理A(n=6):地氟醚预处理20min,10min清洗期,其后处理同2组;组4较长期缺氧,复氧(n=5):细胞缺氧48h,复氧24h;组5地氟醚预处理B(n=5):在较长期缺氧/复氧之前,地氟醚预处理20min,10min清洗期。应用放射配体结合法测各组的M受体密度变化。结果 短期缺氧/复氧可致M受体密度显著降低(与1组比:P<0.01),地氟醚预处理(组3)可显著逆转M受体密度的降低;较长期缺氧/复氧可致M受体密度呈进一步下降(P<0.05),地氟醚预处理(组5)不能有效地逆转其所致的M受体密度的下降。结论 M受体参与了心肌细胞缺氧,复氧损伤的调节;地氟醚预处理的心肌保护作用与缺氧/复氧的时间长短有关。Objective To investigate the effect of desflurane pretreatment on the density of muscarinic (M)-receptors on anoxic/reoxygenated rat cardiac myocytes. Methods Myocytes were obtained from ventricle of 2-3 day SD rats and incubated for 3 days. Then they were randomly allocated to one of 5 groups: group I received no treatment and served as control ( n = 5); group Ⅱ underwent 2 h hypoxia (liquid culture medium was insufflated with a gas mixture of 1 % CO2 , 1 % O2 and 98% N2) followed by 1h reoxygenation ( n = 6) ; group Ⅲ was pre-treated with 1.5 MAC desflurane (9% ) for 20 min, followed by 10 min washout before the same duration of hypoxia/reoxygenation as in group II ( n = 6) ; group IV underwent longer duration of hypoxia (48 h) followed by 24 reoxygenation ( n = 5) ; group V was pre-treated with 1.5 MAC desflurane (9% ) for 20 min followed by 10 min washout, then underwent longer duration of hypoxia/reoxygenation as in group IV ( n = 5) . The density of M-receptors was measured by the radio-ligand binding assay (RLBA) .Results The density of M-receptors was significantly lower in group Ⅱthan that in group I ( P < 0.01) , Desflurane pretreatment effectively attenuated the decrease in the density of M-receptors induced by hypoxia/reoxygenation. The difference in the density of M-receptors was not significant between group II and Ⅳ . Desflurane pre-treatment did not attenuate the decrease in the density of M-receptors induced by longer duration of hypoxia/reoxygenation. Conclusion M-receptors are involved in the regulation of hypoxia/reoxygenation injury. The protective effect of desflurane pre-treatment is related to the duration of hypoxia/reoxygenation.
