摘  要:目的 探讨成骨细胞悬液接种于脱蛋白型松质骨中对骨缺损修复的能力。方法 以 2 8天胎兔颅骨为骨源 ,用酶消化法分离成骨细胞 ,取第 3代成骨细胞制成细胞悬液 ,以 5× 1 0 6 /ml浓度接种于具有三维空间结构的异种脱蛋白型松质骨中 ,然后用此复合物修复兔颅骨缺损 ,本组作为实验组 (共 1 0只 )。对照组Ⅰ (共 1 0只 ) ,仅用异种脱蛋白型松质骨修复兔颅骨缺损。对照组Ⅱ (共 1 0只 )为成骨细胞。对照组Ⅲ (共 1 0只 )为空白对照组。结果  8周后 ,实验组 :X线片均显示骨缺损周围有骨小梁通过 ,支架上有钙化阴影。大体观察 :缺损周围与正常骨组织之间无分界线 ,植入物硬度接近于正常骨组织。组织学分析 :支架上有大量新骨形成 ,支架有部分被降解吸收。四环素荧光标记证实在支架上有新生骨。所有对照组均无新生骨 ,仅见缺损区有纤维样组织覆盖。Objective To study the ability of osteoblast to repair rabbit cranial bone defect after cultured in vitro and then seeded into porous deproteinized bone. Methods The osteoblast was acquired from the cranial bone of rabbit of 28 days old after digesting by enzyme, the third generation of those osteoblasts was seeded into the porous heterogenous deproteinized bone with three dimensional structure at a density of 5×10 6/ml, the formed compound was then used to repair the defect, 10 rabbits were included in this experiment group. The same defect receiving heterogenous porous deproteinized bone or osteoblast, or nothing at all respectively were served as the three control groups, each included ten rabbits. Results After eight weeks bone trabecula was observed in the experiment group to pass the defect by x-ray and calicified shadow was also observed. Gross inspection: no demarcation was resent between the region of bone defect and normal bone, the xenograft was in the consistency similar to normal bone. Histological analysis: plenty of new bone formed on the scaffolds, part of those scaffolds was degraded, the formation of new bone was confirmed by tetracycline fluorescence labelling method. No new bone formed in all control groups, the defect was covered with fibrous tissue only in the control groups. Conclusion Osteoblast cultured in vitro could be used for the repair of bone defect after proliferation and seeded into porous heterogenous deproteinized bone, and could be used as a new way to repair bone defecet clinically.
