体液中苯二氮类药物提取回收率的研究

摘　　要：目的　探讨溶剂类型和pH选择对动物体液中苯二氮 卓艹 类药物提取回收率的影响。方法　将大鼠连续 2d每日注射苯二氮卓类药物 2mg/kg ,2d后每日采其血液和尿液 ,选择不同的试剂 ,缓冲溶液和pH用UV法测定其体液中药物浓度和计算回收率。结果　实验一组和二组、三组大鼠体液中安定、利眠宁和硝基安定以乙醚溶剂和用pH8为缓冲溶液的提取回收率最高。结论　动物体液中苯二氮卓类药物提取回收率决定于提取溶剂的类型和提取时缓冲溶液的pH的选择。Objective The purpose of this study is to explore what influence the type of solvent and pH will exert benzodiazepines recover rate .Methods Inject benzodiazepines 2 mg/kg into wistar white rat for two days, and then, collect its blood and urine everyday , choose different kinds of solvent ,buffer and pH detect its drug concentration with UV method , then calculate the recover rate.Results valium, Librium and chcordiazeproxide extracted with Aether solvent and PH8 buffer have the highest recover rate .Conclusion The recover rates of benzodiazepines in the body fluid were decided by the type of solvent and the pH of buffer.
