摘  要:目的 建立的全血中环孢素A的HPLC测定法。方法 全血经多步处理后采用C18柱 ( 15 0mm× 4 6mm ,10 μm)分离 ;流动相为甲醇 水 ( 80∶2 0 ) ;流量为 1 0ml/min ;检测波长为 2 14nm ;柱温为 6 5℃ ;环孢素D为内标。结果 全血中杂质不干扰样品的测定 ,标准曲线线性关系良好 (r=0 998) ;最低可定量浓度为 5 0ng/ml;高、中、低 3种浓度的日间和日内变异均小于10 0 % ;相对回收率在 95 9~ 10 5 1% ;符合药物生物样品分析要求。Objective To develop a reversed-phase liquid chromatographic method for the determination of cyclosporin-Α concentration in whole blood.Method The whole blood samples was separated by column C 18 (150 mm×4.6 mm,10 μm) after an improved extraction. The mobile phase was methanol-water(80∶20);the flow rate was 1.0 ml/min;wave length of UV detector was at 214 nm. The column temperature was maintained at 65 ℃. The Internal standard was Cyclosporine D(CsD). Results No other constituents in the sample interfered the determination of CyA,and correlation coefficient of performance calibration curve was 0.998,LOQ was 50 ng/ml. Within-day and between-day coefficient of variance of three concentrations were all lower than 10.0%,and the accuracy range was 95.9~105.1%. The method was suitable for TDM.
