摘 要:目的 :研究并优化大孔树脂法分离纯化山茱萸总皂苷 .方法 :70 0mL·L-1乙醇提取后 ,上HPD 3 0 0型大孔吸附树脂 ,水洗后分别用 3 0 0mL·L-1,5 0 0mL·L-1,70 0mL·L-1乙醇洗脱 ,考察大孔树脂富集、纯化山茱萸总皂苷的吸附性能和洗脱参数 ,并与常规溶剂提取法相对照 .结果 :山茱萸总皂苷富集于 5 0 0mL·L-1乙醇洗脱液部分 ,洗脱剂用量为上柱样品液的 8倍 ,洗脱率达 81 8% .大孔树脂法与常规溶剂法提取山茱萸总皂苷的得率分别为 2 4 9% ,2 19% .结论AIM: To optimize the extraction and refinement of total saponins from Cornus officinalis by column chromatography of macroreticular resin. METHODS:Total saponins were extracted with 700 mL·L -1 of ethanol and purified with HPD 300 macroreticular resin and eluted with distilled water, 300 mL·L -1 , 500 mL·L -1 and 700 mL·L -1 of ethanol. Adsorption capability and elution parameters were observed and compared with those extracted by routine solvents. RESULTS: Total saponins from Cornus officinalis enriched in 500 mL·L -1 of ethanol eluting solution. The dosage of elution reagents was 8 times as the sample volume and the elutive ratio was 81 8%. The average gaining rates of the two methods were 2 49% and 2 19% respectively. CONCLUSION: It is a good method to use macroreticular resin to adsorb and purify total saponins in cornus officinalis.
