摘 要:目的:观察人毛乳头细胞体外长期培养的生长特性,为深入阐明毛乳头细胞凝集性生长的分子机制打下基础。方法:采用二步酶消化法分离毛乳头后,于DMEM培养基中培养,观察长期传代培养中毛乳头细胞生长方式的改变,并用计数法绘制不同传代水平细胞的生长曲线。结果:培养中的毛乳头细胞多呈梭形,细胞在融合前出现多层凝集性生长的细胞团,并具有周期性,凝集性生长状态下毛乳头细胞增殖速度加快;凝集性生长随着传代次数增加而减弱,表现为凝集性生长的细胞团数目减少、体积变小。结论:体外培养的人毛乳头细胞具有周期性凝集性生长的特性,凝集性生长的特性可能与其功能密切相关。Objective:To observe the growth character o f long-term cultured human dermal papilla cells and to establish the research b asis in molecular mechanism of aggregative behavior of dermal papilla cells(DPC ).Methods:Dermal papillae were separated by two-step enzymatic digestion and cultured in DMEM medium.The growth character of dermal papilla cells during lo ng-term subculture pro-cedure was observed and the growth curve during differe nt passage of subcultured HPC was analyzed by counting and grafting.Re sults: Cultured dermal papilla cells were similar to fibroblasts in appearance,but ge nerally and periodically exhibited an aggregative pattern in culture,forming sm all clumps of cells and overlapping each other(with other areas of dish remaini ng clear),named as aggregative behav-ior or aggregative growth pattern.DPC w ith aggregative behavior grew more vigorously than those DPC with non-aggregati ve behavior.However,this aggregative ability became less and even disappeared following passage subculture and elongation of culture time.Con-clu sions:Cu ltured dermal papilla cells generally and cyclically develop the characteristic of aggregative behavior in culture,which is closely related to their abilities including induction and control of hair cycle.
