摘 要:对野罂粟细胞悬浮培养及其次生代谢物野罂粟生物碱进行了研究。结果显示 ,在培养的中后期补充营养物质可促进细胞生长及生物碱的累积。 3 %的蔗糖浓度对细胞的生长最为有利 ;4%的蔗糖对总生物碱的累积最为适宜。悬浮细胞在pH 5 .7的培养基中生长最为旺盛 ,而在pH 5 .4的培养基中总生物碱的含量最高。HPLC法测定野罂粟悬浮培养物中野罂粟碱的含量为 0 .0 2 3 5mg gDW ,黑龙辛甲醚的含量为 0 .0 2 2mg gDW。Cell suspension cultures of Papaver nudicaule were o btained and their alkaloids content was determined by HPLC. The biomass of suspension cells and th e yield of alkaloids can be accelerated after the supplement of nutritions in th e middle or late phase of the growth cycle. It has been showed that 3% sucrose w as most favorable to the cell growth and 4% sucrose to the yield of total alkalo ids by the study of the carbone source. The research of the original pH value of medium showed that the biomass of suspension cells got the fastest growth at pH 5.7 in the medium, and the highest yield of total alkaloid at pH 5.4. The amoun t of nudicauline in the cell by suspension culture was 0.0235 mg/g DW and the a muresinine was 0.022mg/g DW respectively by means of HPLC.
