摘 要:目的 :观察天麻注射液对小鼠脾淋巴细胞转化的影响。方法 :采用 3H_TdR参入法检测ConA刺激的小鼠脾淋巴细胞增殖。结果 :①腹腔注射2 5mg/kg,5mg/kg,10mg/kg,20mg/kg天麻注射液12h,ConA刺激的小鼠脾淋巴细胞转化明显高于对照组 (P<0.05,P<0.01) ;40mg/kg的天麻注射液对小鼠脾淋巴细胞转化无明显影响。②体外实验0 1μg/ml~100μg/ml的天麻注射液组与对照组比较 ,小鼠脾淋巴细胞转化明显增加 (P<0.05,P<0.01) ;200μg/ml~400μg/ml天麻注射液组的小鼠脾淋巴细胞转化与对照组比较 ,差异无显著性意义 ;800μg/ml天麻注射液组的小鼠脾淋巴细胞转化则显著低于对照组。结论 :低浓度的天麻注射液(腹腔注射2 5~20mg/kg,体外实验0 1~100μg/ml)具有提高小鼠脾淋巴细胞转化的功能 ,中等浓度的天麻注射液(腹腔注射40mg/kg,体外实验200~400μg/ml)对ConA刺激的小鼠脾淋巴细胞转化无明显作用 ,而高浓度的天麻注射液(体外实验800μg/ml)Purpose: To study the effects of Tianma Injection on the mouse's splenic lymphocyte proliferation in vivo and vitro. Methods: The proliferation of the ConA_stimulated splenic lymphocyte was measured with incorporation of 3H_methylthymidine( 3H_TdR) in mice. Results: Intra_abdominal injection of Tianma Injection ( 2.5 mg/kg,5mg/kg,10mg/kg and 20mg/kg) improved the splenic lymphocyte transformation of the ConA_stimulated mice. However, the studies showed the inhibition of the splenic lymphocyte transformation by 40mg/kg of Tianma Injection. In vitro, lower doses (0.05~100μg/ml ) of Tianam Injection helped an increased rate of 3H_TdR_uptakes whereas responsiveness of splenic lymphocytes to higher doses(200~400μg/ml) was not modified. Contrary to the splenocyte responses, activation by Tianma Injection (800μg/ml) was characterized by a decreases proliferation, when compared to the background count of unstimulated cells. Conclusions: The findings prove that lower doses of Tianma Injection can enhance certain parametere of splenic lymphocyte function in vivo and in vitro while higher doses suppress it.
