摘  要:目的 研究青春期小鼠Leydig细胞中转醛酶活性、蛋白表达水平与睾酮生成的相关性。 方法 原代培养Leydig细胞 ,加入HCG刺激 ,酶联免疫法测定培养 2、8小时细胞上清睾酮浓度 ,同时测定转醛酶活性 ,Westernblot检测转醛酶蛋白表达水平。结果  (1)刺激组Leydig细胞上清睾酮浓度明显高于对照组 ,组间存在显著性差异 (2小时P <0 .0 1,8小时P <0 .0 0 1)。 (2 )刺激组Leydig细胞转醛酶活性明显高于对照组 ,组间存在显著性差异(2小时P <0 .0 5 ,8小时P <0 .0 0 1)。 (3)Leydig细胞刺激组和对照组间转醛酶蛋白表达水平无显著差异 (P >0 .0 5 )。结论 HCG促进小鼠Leydig细胞生成睾酮的同时 ,转醛酶活性同步提高 。Objective To study the relationship between transaldolase activity,protein expression and testosterone synthesis in puberty mouse leydig cells.Methods After HCG were added to stimulated groups,Leydig cells were cultured in vitro for 2 to 8 hours,testosterone concentration were measured by Enzyme linked immunoadsordent assay (ELISA),meanwhile transaldolase activities were measured and protein expression were measured by Western blot.Results (1) After cultivation testosterone concentration in stimulated groups were both significantly higher than in control groups (2 hours P <0.01,8 hours P <0.001).(2) After cultivation transaldolase activities in stimulated groups were both significantly higher than in control groups (2 hours P <0.05,8 hours P <0.001).(3) After cultivation the ratio of A isoforms,B isoforms and total transaldolase protein to β actin in stimulated groups had little difference with control groups.Conclusions When HCG stimulated testosterone synthesis in leydig cells,transaldolase activities increased at the same time,which verified the positive relationship between transaldolase activities and testosterone synthesis in puberty mouse Leydig cells.
