摘 要:目的 研究不同剂量丙泊酚对自发性高血压大鼠 (SHR)心肌 β 肾上腺素能受体 (β AR)、腺苷酸环化酶 (AC)及血浆心钠素 (ANP)和神经肽Y(NPY)的影响 ,探讨丙泊酚对SHR心血管影响的机制 ,指导临床麻醉高血压病人的合理用药。方法 SHR2 4只 ,随机分为三组。空白对照组(Ⅰ组 ) ;丙泊酚低剂量组 (Ⅱ组 )和丙泊酚高剂量组 (Ⅲ组 )。观察麻醉期间血压、心率、β AR、cAMP(AC活性用cAMP间接反映 )、ANP和NPY。结果 丙泊酚麻醉使血压明显下降 (P <0 0 5 ) ,心率不变 ,Ⅱ组Bmax值降低 ,Ⅲ组Bmax和KD值均降低 (P <0 0 5 ) ;且 β1 比例、AC活性和NPY值明显降低 (P <0 0 5 ,P <0 0 1 ) ,Ⅱ、Ⅲ组比较差异显著 (P <0 0 5 ) ,ANP无明显变化。结论 丙泊酚明显影响SHR心脏β AR/AC系统和NPY的分泌 ,并与剂量相关 。Objective To study the alteration of cardiac beta adrenergic receptors and their subtypes,adenylate cyclate(AC) plasma neuropeptede Y and atrial natriuvetic polypeptide(ANP)in spontaneously hypertensive rats(SHR) with different dose of propofol.Method 24SHR were devided randomly into three groups:control group (Ⅰ),low dose propofol(5mg/kg)(Ⅱ)and high dose propofol(12mg/kg)(Ⅲ).The BP,HR β AR and their subtypes,AC,ANP,NPY were determined.The activity of AC was indirectly reflected by cAMP.Results The arteriah blood pressure was significantly decreased in group Ⅱ and Ⅲ,but heart rate was not changed.The activity of AC,Bmax,NPY decreased( P< 0 05) in Ⅱ,Ⅲ,but KD was not changed.The ratio of β1 decreased( P< 0 05).ANP was not changed in Ⅱ,Ⅲ.Conclusion Propofol depresses cardiovascular function in a dose dependent mamner partly because of the decrease in the secretier of β AR/AC system and the NPY.
