改良PAGE法用于人类珠蛋白多态性研究

摘　　要：目的 :探讨人类新生儿HbF中珠蛋白链简便、快速、准确的分离方法。方法 :对传统的含TritonX 10 0的酸性聚丙烯酰胺凝胶电泳法的凝胶系统作了进一步的改进 ,其中TritonX 10 0和TEMED等主要物质的浓度作了调整 ,不同于其它文献报导的。结果 :该改良法能使凝胶聚合时间缩短、一次电泳时间从 6h缩短为 2h ,HbF中各珠蛋白链得到良好分离 ,电泳条带清晰 ,无拖尾现象 ;结论 :该方法操作简便 ,分离效果好 ,电泳时间缩短 ,极大地提高了研究效率。Objective:To seek a method for separating human globin chains in HbF in the new born babies.Methods:Concentration of the main matters(Triton X 100 and TEMED)in the polyacrylamide system in traditional polyacrylamide gel electrophoresis(PAGE)was modified.Results: The globin chains can be well separated by this modified method and the electrophoresis time has been shorted down (from 6 hours to 2 hours ).Conclusion:This method is not only seperating the globin chains well, but also easy to operate and shortens the time of electrophoresis,So it greatly increases the research efficiency in this field.
