摘  要:目的:探讨人体血管平滑肌细胞(Vascular smooth muscle cells,VSMC)的一氧化氮合酶(Nitric oxide synthase,NOS)—一氧化氮(Nitric oxide,NO)系统的变化特点及其与高血压病(Essential hypertension,EH)发病的关系。方法:对EH患者和血压正常者(NT)的动脉VSMC进行培养,分别测定EH组和NT组的NO含量和NOS活性,观察其动脉VSMC分泌NO和NOS的特点。结果:(1)EH组VSMC的N0含量和NOS活性分别为(42.73±6.%)μmol/(2.5×^6cells)和(0.24±0.05)nmol/(2.5×10^6cells),均显著低于NT组(74.52±4.73)μmol/(2.5×10^6cells)和(0.55±0.10)nmol/(2.5×10^6cells)(P<0.01);(2)EH组和NT组VSMC的N0含量和NOS活性随培养时间延长均显著增加和增强(P<0.05);(3)EH组和NT组的NO含量随NOS活性增强而增加,并呈显著正相关(P<0.01)。结论:ET患者的NOS—N0系统可能存在功能和结构的异常,并参与EH的发生和发展。Objective To investigate the changes of nitric oxide synthase (NOS) - nitric oxide (NO) system in vascular smooth muscle cells (VSMC) of patients with essential hypertension (EH), and role of these changes in the pathogenesis of hypertension. Methods The VSMC of EH patients and normotensives (NT) were cultured. The NO contents and NOS activities of the VSMC in the EH and NT groups were measured, and the characteristics of NO and NOS secretion from VSMC were observed. Results (1) The NO content and NOS activity of VSMC in the EH group were (42.73±6.76) μmol/(2.5×10~6 cells) and (0.24±0.05) nmol/(2.5×10~6 cells) respectively,significantly lower than (74.52± 4.73) μmol/(2.5 ×10~6 cells) and (0.55±0.10) nmol/(2.5 ×10~6 cells) in the NT group ( P < 0.01). (2)The NO concentration and activity of NOS in VSMC of both groups increased with increasing cultivation time. (3)The concentration of NO in the two groups increased with the increasing activity of NOS,and there was a positive correlation between the NO concentration and the NOS activity. Conclusions The NOS-NO system in patients with EH is probably abnormal in structure or function,and is involved in the production and development of essential hypertension.
