异体耳廓移植后局部淋巴结MIP-3β的含量变化及趋化活性的研究

摘　　要：目的　研究异体耳廓移植后局部淋巴结内趋化因子MIP 3 β的含量变化及对树突状细胞的趋化作用。 方法　进行吻合血管的大鼠同种异体耳廓移植 ,ELISA法测定术后局部淋巴结MIP 3 β的含量变化趋势 ,应用改良Boyden小室进行淋巴结组织液对体外培养树突状细胞的趋化实验。结果　同系移植组术后局部淋巴结MIP 3 β含量升高 ,持续至术后第 2天 ,以后恢复至正常水平 ;异系移植组MIP 3 β含量持续上升 ,后期显著高于正常淋巴结内的表达水平 ,其对体外培养树突状细胞的趋化作用增强 ,这种趋化作用可被MIP 3 β抗体所阻断。 结论　器官移植后局部引流淋巴结内MIP 3 β表达增高 ,促进树突状细胞的迁移运动 。Objective To study the alteration of MIP 3β content in draining lymph nodes after auricle allotransplantation and their chemotaxis to dendritic cell. Methods The auricles were transplanted from BN to Lewis rats under microsurgery. The content alteration of MIP 3β in head and neck draining lymph nodes was evaluated by ELISA and their chemotaxis to dendritic cell was detected in modified Boyden chamber. Results The content of MIP 3β in DLNs of isograft groups increased within 2 d after operation and then recovered to normal level while that of allograft displayed the increasing trend continuously and their homogenate showed enhanced chemotaxis to cultured DC in vitro , which could be blocked by MIP 3β antibody. Conclusion The expression of MIP 3β in DLN is enhanced after allotransplantation, which accelerates DC migration and antigen presence. It may be one of the reasons to induce and promote allo antigen presence and rejection response.
