摘  要:目的 :探讨人小细胞肺癌细胞株NCI H4 4 6的生长抑素受体体外调变的规律 .方法 :采用体外放射配基结合分析法 ,以12 5I标记的生长抑素类似物 (RC 16 0 )为配基 ,在与NCI H4 4 6细胞进行体外饱和及竞争结合实验的基础上 ,进行RC 16 0诱导下NCI H4 4 6细胞株的生长抑素受体内化及上调实验 .结果 :温育 15min时 ,内化组与对照组的内化率分别为 (2 2 .3± 2 .5 ) %和 (8.2± 3.3) % (P <0 .0 1) ,膜结合率分别为 (4 3.8± 4 .0 ) %和 (12 .4± 1.6 ) % (P <0 .0 1) .低浓度的非标记RC 16 0暴露前 13h上调率增加缓慢 ,仅为 (2 0 .0±1.3) % ,其后上调率迅速增加 ,并于暴露后 19h达到顶峰(76 .5± 2 .6 ) % .结论 :RC 16 0可使NCI H4 4 6细胞表面生长抑素受体内化并上调 。AIM: To investigate the in vitro modulation of somatostatin receptor (SSTR) on the surface of small cell lung cancer cells, NCI H446. METHODS: By in vitro saturable binding study and competitive binding study, combined with radio ligand binding assay, RC 160, the somatostatin analog, was used to induce SSTR internalization and upregulation. RESULTS: 15 min after initial incubation, the internalization rates in experimental group and control group were (22.3±2.5)% and (8.2±3.3)%, and the membrane binding rates were (43.8±4.0)% and (12.4±1.6)%, respectively. There was statistical significance between the two groups( P <0.01). 13th hours after exposure to low concentration of non labeled RC 160, the upregulation rate was (20.0±1.3)%, showing a mild increase. The upregulation rate increased rapidly and reached the climax (76.5±2.6)% at the 19th hour, and then decreased gradually. CONCLUSION: RC 160 may modulate the internalization and upregulation of SSTR on the surface of NCI H446 cells, which has a time phase correlation.
