摘 要:目的观察尼莫地平和钙剂对快速老化(SAMP6)小鼠血清及各脏器Ca2+含量的影响作用,探讨“Ca2+重新分布假说”与老年性骨质疏松形成的关系。方法用原子吸收法测定服用尼莫地平或凯思立D的SAMP6小鼠血清、股骨、心、肺、肝、脑、肾、肌肉和皮肤中Ca2+含量,并与其自身空白对照及正常老化小鼠SAMR1作对照。结果服用尼莫地平和钙剂(凯思立D)的SAMP6小鼠血清(P<0.01),股骨(P<0.01)和皮肤(P<0.01或P<0.05)中Ca2+含量明显高于SAMP6空白对照组;尼莫地平组SAMP6小鼠肾脏中Ca2+含量明显低于其他各组(P<0.01);脑、肝、心脏等脏器中Ca2+含量没有显著性差异。结论钙剂(凯思立D)和钙离子拮抗剂(尼莫地平)均可提高血清及骨质中Ca2+含量。Ca2+体内重新分布可能与SAMP6小鼠骨质疏松的形成有一定关系。钙离子拮抗剂尼莫地平对老年性骨质疏松症可能存在治疗作用。Objective To observe the effects of Nimodipine and Calcichew - D3 on the Calcium concentration and distribution of the blood and viscera of SAMP6 mice and discuss the hypothesis, the fundamental mechanism in senile osteoporosis might be caused by calcium redistribution. Methods AA - 680 atomic absorption/flame emission spectrophotometer(SHIMADZU) was used to assay calcium concentration of the blood and viscera of SAMP6 mice which were given Nimodipine of Calcichew - D3 for 50 days. SAMR1 mice that were normal aging and SAMP6 without any treatments were used as control. Results Calcium concentration was significantly decreased in the blood serum and the femur of the SAMP6 control group mice, compared with that of the SAMR1 control group mice ( P = 0.006 and P - 0.004, respectively) . Calcium concentration of blood serum was significantly increased in the Nimodipine group ( P < 0.01) and in the Calcichew - D3 group ( P < 0.01)as compared with the SAMP6 control group. In the Nimodipine group ( P <0.01) and the Calcichew- Dj group ( P < 0.01) ,calcium concentration of femur was significantly increased, compared with the SAMP6 control group. Calcium concentration of the kidney was significantly decreased in the Nimodipine group compared with other groups ( P < 0.01). There is a significantly decrease in calcium concentration of skin in the SAMP6 control group,compared with the Calcichew-D3 group and the Nimodipine group ( P = 0.010 and P = 0.001 respectively) There is no significant difference in calcium concentrations of liver,brain and heart in groups.Conclusions Calcium supplement (Calcichew-D3) and calcium antagonist (Nimodipine) can both increase the blood and the bone calcium concentration. Osteoporosis in SAMP6 could be explained by the hypothesis about the distribution of calcium concentration in some degree. It suggested that Nimodipine might have a therapeutical effect on senile osteoporosis.
