摘 要:目的 :观察不同时限单侧输尿管梗阻 (UUO)模型大鼠肾皮质过氧化物酶体增生物激活受体 (PPAR)γ的表达变化及其与巨噬细胞浸润的关系。 方法 :用免疫组化、RT PCR以及Westernblotting的方法检测UUO大鼠梗阻3天、7天、14天的PPARγ在核酸水平和蛋白质水平的表达量变化以及表达位置的改变 ,其与ED 1阳性巨噬细胞浸润数的关系。 结果 :假手术对照组PPARγ主要表达于肾脏内髓集合管 ,而UUO状态下肾小管上皮细胞出现PPARγ的表达。半定量分析显示UUO后 3天PPARγ的表达开始有增高趋势 ,7天时PPARγ的mRNA和蛋白质水平达到峰值(与对照组相比分别为 3 33倍和 4 36倍 ) ,而梗阻后 14天其表达已开始下降。病理学和免疫组化结果显示 ,UUO 7天时巨噬细胞浸润明显 ,但纤维化改变尚不显著。UUO 14天时炎性细胞积聚明显 ,但巨噬细胞浸润数下降 ,出现明显的小管萎缩和间质纤维化。UUO后PPARγ的表达与巨噬细胞浸润显著相关。 结论 :UUO模型大鼠肾皮质中PPARγ的表达量增加 ,并在肾小管部位出现PPARγ的表达。Objective:Tubulointerstitial fibrosis is the common results of progressive renal disease. Peroxisome proliferators activated receptors gamma (PPARγ) has been reported to have anti inflamative and anti fibrotic effects as they are usually increased in tissue injury. These effects have not ever been investigated in obstructive renal injuries. In this study, we studied the expression of PPARγ at different periods of rat unilateral ureteral obstruction(UUO) model, a model of progressive renal fibrosis, and analyzed the relationship between renal cortex PPARγ expression and macrophage infiltration. Methodology:Rats were sacrificed after 3 days, 7days and 14 days of UUO, and sham operated rats were sacrificed after14 day as the control (CTL). Immunohistochemical staining , RT PCR and Western blotting were applied to study the renal tissue PPARγ expression. Renal cortex PPARγ mRNA and protein levels were compared at different time points after unilateral ureteral obstruction. At the same time, infiltrated ED 1 positive macrophage was observed by immunohistochemical staining. Results:Detectable basic PPARγ expression was observed in sham operated control rats in renal inner medulla, mainly concentrated at collecting tubules. In UUO rats, PPARγ expression was found increased and extended to renal tubular epithelial cells. Increased PPARγ mRNA expression was found after 3 days of UUO, and significantly increased PPARγ mRNA expression was found 7days after UUO ( P <0 001),and the increased expression of PPARγ mRNA was found to be down regulated at the 14th day after UUO. Increased PPARγ protein expression has the same pattern of changes as that of PPARγ mRNA expression, with the 7th day's peak value 4.36 times that of the control group ( P <0 05) and still kept significantly increased at the 14th day of UUO (2 05 times compared with the control group, P <0 05).
