摘  要:作者用醋酸纤维素薄膜作载体,建立了一种新颖的、快速的、简单实用的微菌落技术,即将细菌接种于膜,培养3~6小时后,透明、固定、染色,观察微菌落形态或进行微菌落计数。使用该技术,对139株细菌的微菌落进行观察,积累了大量微菌落资料;借助微菌落特征,对72株葡萄球菌中的金黄色葡萄球菌进行鉴定,与常规法的符合率为90.28%;并首次将计数微菌落的方法用于临床尿液标本细菌的快速定量测定,与常规法比较,具有明显的优越性。A new rapid and usefulmicrocolony technique was developed byusing cellulose-acetate membrane as asupporter. The bacteria were inoculatedon the membrane and cultivated for 3-6h.Then the membrane was rendered, fixedand colored. The microcolonies were ob-served and counted under the microscope.By using this technique the microcoloniesof 139 strains of bacteria were observed.72 strains of Staphylococcus weredifferentiated by means of observing thecharacteristics of the microcolonies. Thecorrelation rate between the microcolonymethod and the routine method was90.28%. This technique was also first usedto determine the bacteria numbers inurine within 6h, and the advantages of itwere significant compared with the platecount method.
