高效液相色谱法测定康欣胶囊大黄素的含量

摘　　要：目的:建立反相高效液相色谱法测定康欣胶囊中大黄素的含量。方法:取药粉1g盐酸水解后,用氯仿提取总大黄素,以十八烷基硅烷键合硅胶为填充剂;甲醇—水为流动相,作梯度洗脱,10min内甲醇浓度自100％降为53％(V/V),检测波长为290nm。结果:大黄素在3.6μg/ml～19.0μg/ml范围内线性关系良好,r=0.9997;样品平均加样回收率为99.32％(n=5),RSD为1.62％;重现性试验RSD为1.83％(n=5)。结论:该方法灵敏度高,专属性可靠,重复性好,可作为康欣胶囊的含量测定方法。Objective:To establish a RP-HPLC method for the determination of emodin in KangXin Capsules. Method:The determination was carried out on C18ODS column with mobile phase of methanol-water and detection wavelength at 290nm according to the external standard method Results;The emodin sample size showed a good linear relationship at the range of 3. 6ug/ml^19. 0ug/ml and correlation coefficient was 0. 9997. The average recovery of the added sample was 99. 32%(n = 5) and RSD was 1. 62%. The RSD in a duplicate test was 1. 83%(n = 5). Conclusion :The method is simple,accurate .reproducible and can be used for content determination of emodin of KangXin Capsules.
