摘 要:目的 :观察正常大鼠、肥胖大鼠和复方大黄制剂灌服 1个月肥胖大鼠脂肪细胞瘦素和C/EBPαmRNA表达的变化及C/EBPα对瘦素表达的调控。方法 :用大、中、小剂量分组 (2 0mg·10 0g-1;4 0mg·10 0g-1;12 0mg·10 0g-1)对肥胖大鼠进行复方大黄制剂灌服。制备脂肪细胞悬液后采用免疫组化ABC法及WesternBlot检测脂肪细胞瘦素表达水平。应用定量RT PCR技术检测脂肪细胞中C/EBPα基因mRNA表达的变化。结果 :肥胖大鼠灌服中剂量复方大黄制剂 30d后体重明显减轻 ;Lee’s指数明显降低。免疫组化ABC法及WesternBlot检测瘦素表达水平 ,发现脂肪细胞瘦素表达明显减弱。定量RT PCR结果发现 ,肥胖大鼠脂肪细胞中C EBPαmRNA表达水平较正常大鼠高 ,在中剂量复方大黄制剂灌服 1个月后 ,随着肥胖大鼠体重的减轻 ,脂肪细胞中C/EBPαmRNA水平也随之降低 ,与瘦素水平降低程度呈显著正相关 (r =0 98,P <0 0 1)。结论 :肥胖大鼠脂肪细胞的瘦素表达水平较正常大鼠高 ;灌服复方大黄制剂减肥有效的同时 ,脂肪细胞瘦素表达水平明显下降。肥胖大鼠脂肪细胞中C/EBPαmRNA表达水平在复方大黄制剂治疗后明显降低 ;其降低程度与脂肪细胞瘦素表达水平间呈显著正相关。齌o elucidate the changes of leptin expression in obese fats and regulatlon of C/EBP α on leptin expression 1 month after oral administration of complex prescription of rhubarb. Methods: Obese rats were treated with different dosage (20mg, 40mg,120mg/100g B.W/d) for a month. Immunohistological (ABC) method and Western Blot analysis of leptin in celiac adipocytes were performed. RT/PCR technique was used to quantitate the levels of C/EBP α mRNA expression in adipocytes. Results:After oral administration for a month, the weight loss effect appeared and the Lee's index decreased dramatically in obse rats. The expression of leptin in adipocytes lowered. The expression of C/EBP α mRNA in adipocytes of obese rats was stronger than that in the control group. With the treatment of complex precresption of rhubarb, the expression of C/EBP α mRNA lowered. There was a positive relationship between C/EBP α mRNA expression and leptin expression. Conclusions: High expression of leptin and C/EBP α mRNA appears in adipocytes of obese rats. After oral administration of complex prescresption of rhubarb, the levels of leptin and C/EBP α expression declines. There is a close relationship between them. It indicates that the increase of leptin expression in adipocytes is regulated by C/EBP α to some extent in obese rats.
