摘 要:目的 :观察弹性蛋白酶气管内滴入复制大鼠肺气肿模型的可靠性。方法 :Wistar大鼠 3 0只随机分为两组 ,正常对照组和模型组 ,每组各 15只。模型组采用弹性蛋白酶气管内滴的方法复制实验性肺气肿模型 ,2 5天后观察各组大鼠的体重、肺体积和病理学变化 ,免疫组化法观察增殖细胞核抗原 (PCNA)的表达情况。结果 :模型组体重和肺体积分别为 ( 3 3 6± 16) g、( 5 .1± 0 .6)cm3,与对照组 ( 3 5 2± 15 ) g、( 4.3± 0 .4)cm3比较差异有显著性 (P<0 .0 1) ;模型组的平均肺泡面积和平均内衬间隔分别为 ( 3 745± 679) μm2 和 ( 5 5 .0 3± 8.2 3 ) μm ,与正常对照组( 195 4± 3 44 ) μm2 、( 42 .0 2± 5 .98) μm比较差异有显著性 (P <0 .0 1) ;PCNA指数对照组为 ( 2 0± 4) ,模型组为 ( 5 7± 6) ,(P <0 .0 1) ,差异显著。结论Objective: To observe the reliability of elastase in inducing experimental emphysema in rats. Methods: Thirty Wistar rats were randomly divided into two groups, the normal group(A), the model group(B), fifteen rats in each group. Experimental emphysema model were made by instilling elastase into rat lungs of Group B. After twenty-five days, body weight, volume of the lung and pathological changes of rats were observed. The proliferating cell nuclear antigen (PCNA) was examined by immunohistochemical method. Results: Compared with rats of group A, there was significant difference in rats of group B in body weight and volume of the lung \[(336±16)g vs(352±15)g\], \[(5.1±0.6)cm 3 vs (4.3±0.4)cm 3\],respectively (P<0.01). Compared with rats of group A, there was significant difference in rats of group B in mean alveoli area(MAA)and mean lung interval (MLI) \[(3 745±679)μm 2 vs (1 954±344)μm 2,(55.03±8.23)μm vs (42.02±5.98)μm, respectively (P<0.01)\]. In comparison with rats of group A, the PCNA index staining in rats of group B (20±4 vs 57±6) was increased (P<0.01). Conclusion: Instilling elastase into rat lungs can inducing experimental emphysema in rats reliably.
