摘 要:目的 探讨染不同浓度煤尘的肺泡巨噬细胞 (染煤尘 -AM )上清液作用成纤维细胞不同时点对肌醇 - 1,4 -二磷酸 (IP2 )和肌醇 - 1,4 ,5 -三磷酸 (IP3 )信号物质的影响。方法 应用细胞培养技术 ,用小鼠胸腺细胞增殖法和放射免疫分析法 ,分别测定染煤尘 -AM上清液中白细胞介素 1(IL 1)、前列腺素E2(PGE2 )的含量 ;用Dowex - 1× 8阴离子交换柱层析法 ,分析染煤尘 -AM上清液作用于3 H -肌醇标记成纤维细胞后IP2 和IP3 的液闪计数值 (cpm)。 结果 AM培养上清液中IL 1,PGE2 含量随染煤尘浓度的增加而增高 ,与对照组比较差异有显著性。用这种含细胞因子的染煤尘 -AM上清液作用成纤维细胞 ,在不同的作用时点 ,对IP2 的影响不同 ,在 10 ,2 0s时点 ,上清液中IL 1和PGE2 对IP2 显示有上调作用 ,随着作用时间的延长 ,IP2 出现了下调现象。对IP3 的影响表现为 ,在不同的作用时点均随上清液中细胞因子的增加IP3 呈现增加趋势。煤尘组与对照组比较 ,在不同的观察时点显示有不同的差异显著性。结论 染煤尘-AM培养上清液中含有高于对照组的IL 1和PGE2 细胞因子 ,用这种上清液作用于成纤维细胞后 ,可影响成纤维细胞中磷酸肌醇信号物质的含量 ,影响程度与细胞因子的浓度和作用时间有关。Objective To explore effects of supernatant of coal dust exposed alveoli macrophage(AM)at different doses on inositol 1,4 diphosphate(IP 2)and inositol 1,4,5 triphosphate(IP 3)of fibroblast.Methods The level of IL 1 and PGE 2 in culture medium was measured by thymus gland cell proliferation of mice and radio immunity assay.The cpm values of IP 2 and IP 3 of fibroblast treated with coal dust exposed AM supernatant were measured with the method of Dowex-1×8 ion exchange chromatography.Results Compared with the control group,the contents of IL 1 and PGE 2 in this supernatant were statistically increased with the dose of coal dust.When coal dust exposed AM supernatant acted on fibroblast,it had different effects on the levels of IP 2 at different time point,IL 1 and PGE 2 in supernatant made the IP 2 level to increase at 10 s and 20 s,but tended to decrease as time prolonged.For IP 3,it tended to increase as the increase of IL 1 and PGE 2 at different time point.There existed significant statistical difference for IP 2 and IP 3 at different time point between the coal dust group and the control group.Conclusions The level of IL 1 and PGE 2 in coal dust exposed AM supernatant is higher than that of the control group.Phosphate inositol level in fibroblast can be influenced when fibroblast is treated by this supernatant,and the effect might be related with time and the level of IL 1 and PGE 2.
