人参二醇组皂甙促进受损的培养肾小管细胞增殖的作用及机制研究

摘　　要：目的 :观察人参二醇组皂甙 (PDS)对正常培养和经急性缺血再灌注肾损伤家兔血清处理后的小管细胞增殖的影响 ,并探讨其机制。方法 :用夹闭法致家兔肾缺血 ,并提取缺血血清和对照血清 ,分别与人肾小管细胞共培养 ;四甲基偶氮唑盐 (MTT)法测定细胞株吸光度 (A)值以判定其增殖活性 ;苔盼蓝染色计算活细胞率。结果 :1 0 mg/ L的 PDS组 A值 (0 .6 2 76± 0 .0 392 )明显高于对照组 (0 .5 0 75± 0 .0 5 97) ,P<0 .0 5 ;缺血血清组 A值 (0 .1 989± 0 .0 31 3)与活细胞率 (4 6 .0 % )均明显低于其他组 (0 .6 5 94～ 0 .785 0 ,91 .5 %～ 98.9% ) ;PDS加缺血血清组的 A值 (0 .6 5 94± 0 .1 6 78)与对照组 (0 .735 0± 0 .0 81 2 )比较差异无统计学意义。结论 :适量的Objective: To study the effects and mechanisms of panaxadiol saponins promoting the proliferation of normally cultured human renal tubular cells and the cells which suffered by the rabbit serum of acute renal ischemicreperfusion. Methods: The bilateral renal arteries of rabbits were occluded then reperfusion, and the serum(SIR) was acquired. The rabbit serum of control (SC)were acquired in the same time. The proliferative activity of cells were measured with the method of MTT. And the rate of living cells were counted. Results: The value of optical density(OD, as A ) in 10 mg/L PDS group (0 627 6±0 039 2) was significant higher than that in control groups (0 507 5±0 059 7), P <0 05. The value of A and the rate of living cells in SIR group (0 198 9±0 031 3 and 46 0%) were significant lower than those in other groups (0 659 4-0 785 0 and 91 5%-98 9%). But there were not significant differences in the proliferative activity of PDS +SIR groups (0 659 4±0 167 8) and control group (0 735 0±0 081 2). Conclusion: Moderate panaxadiol saponins could accelerate the proliferation of normally cultured human renal tubular cells and the cells which suffered by the rabbit serum of acute renal ischemicreperfusion.
