木贼对动脉粥样硬化早期大鼠内皮细胞凋亡及Bax、Bcl-2表达的影响

摘　　要：目的　观察木贼对大鼠动脉粥样硬化早期血管内皮细胞凋亡及 bax、bcl- 2表达的影响。方法　高脂饲料复制 SD大鼠高脂血症和早期动脉粥样硬化模型 ,同时给以不同剂量木贼水煎剂灌胃 ,实验 1 0 w后 ,流式细胞术定量检测各组主动脉内皮细胞凋亡率及 bax、bcl- 2基因蛋白表达。结果　模型组内皮细胞凋亡率及两个基因表达明显高于正常组 (P<0 .0 1 ) ;木贼组内皮细胞凋亡率与 bax表达量显著低于模型组 (P<0 .0 1 ) ,bcl- 2比较虽无统计学意义 ,但用药后 bax/bcl- 2比值差异具有显著性。结论　木贼可从基因水平调控早期动脉粥样硬化主动脉内皮细胞凋亡 ,干预动脉粥样硬化始动环节及其发生。Objective To investigate the effect of Equisetum that controlling the apoptosis of aorta endothelium by regulating the expression of Bax and Bcl-2 on experimental hypercholesterolemia and atherosclerosis.Methods Forty-eight Sprague-Dawley rats were randomly divided into four groups: a normal group with standard chow diet, and three other groups fed with a high fat diet including two groups plus Equisetum at different doses. The rate of apoptosis and the expressions of Bax and Bcl-2 in aorta endothelium were detected by the way of flow cytometry after ten weeks.Results The rate of apoptosis of endothelium in model group was higher than those in normal group (P<0.01). The expressions of Bax were enhanced (P<0.01). The rate of apoptosis ?the expression of Bax and the amount of Bax/Bcl-2 in Equisetum groups were decreased compared with those in model group (P<0.01).Conclusions Equisetum appear action on regulating the apoptosis of aorta endothelium by regulating the expression of Bax .
