摘  要:为探讨原发性高血压 (EH)肾损害与胰岛素受体 (INSR)基因第 8外显子NsiⅠ酶切多态性的关系 ,采用生化法、多聚酶链反应 (PCR)和限制性内切酶片段长度多态性分析 (RFLP)等技术 ,检测了N2 等位基因在EH肾功能正常 (EH NRF)组、EH肾损害尿毒症 (EH CRF)组及正常对照 (NC)组中的表达。结果显示 ,NsiⅠ多态性与患者的年龄、体重指数 (BMI)、总胆固醇 (TC)及甘油三脂 (TG)等无相关性 ;N2 等位基因频率在EH NRF组及EH CRF组均明显高于NC组 (P <0 0 5、<0 0 1) ,且在EH CRF组高于EH NRF组。提示N2 等位基因可能是中国人EH(特别是EH CRF)To investigate the relation between polymorphism of insulin receptor (INSR) gene exon 8 and hypertensive nephrosclerosis,polymerase chain relation (PCR) and restriction fragment length polymorphism (RFLP) combined with biochemistry method were used to measure the frequency of N 2 allele of INSR gene in normal renal function of essential hypertension (EH-NRF),renal failure of hypertensive nephrosclerosis (EH-CRF) and normal control (NC) subjects.Results showed that there was no relationship between polymorphism of NsiⅠ and the age,body mass index (BMI),serum lipid level.The N 2 allele frequency of INSR gene in EH-NRF and EH-CRF groups were higher than that in NC group(P<0.05,<0.01),and the N 2 allele frequency of INSR gene in EH-CRF group was also higher than that in EH-NRF group.These suggest N 2 allele is a significantly sensitive gene of Chinese EH and EH-CRF.
