摘 要:目的 使用小鼠的卵丘细胞为供核 ,去核的成熟卵浆为受体进行卵子重构 ,观察受精、胚胎发育情况。 方法 昆明小鼠超排卵后获取生殖泡期卵子 ,进行体外成熟。成熟卵子去核后将卵丘细胞核与精子直接注入 ,观察其受精和胚胎发育情况。 结果 卵子去核存活率为 6 9.2 % ( 2 5 8/373) ,注核存活率为 4 4 .1% ( 112 / 2 5 4 )。 2 0 .9% ( 19/ 91)重构卵子排出 1个极体并形成两原核 ,卵裂率为 78.9% ( 15 / 19)。 15个重构卵子于 D2 分裂至 2~ 3个细胞后阻滞 ,无囊胚形成。 结论 将小鼠的卵丘细胞核移植到成熟的去核卵浆中 ,同时注射精子进行激活 ,可以形成重构卵子并受精 ,但出现 2细胞阻滞 。Objective To observe the fertilization and development of reconstructed oocyte by transferring mice cumulus cell into enucleated metaphase Ⅱ oocytes. Methods Germinal vesicle oocytes were collected from stimulated Kunming mice ovary and cultured for in vitro maturation.The nuclei of cumulus cell and sperm were injected directly into the enucleated oocytes.The fertilization and develonpment were obseved. Results The survival rate after enucleation was 69.2 %(258/373),after injection was 44.1 % (112/254).1 polar body was extruded and 2 pronuclear formed in 20.9 % (19/91)reconstructed oocytes.The cleavage rate was 78.9 % (15/19).Embryos blocked at 2 cell stage,no blastocyst formed. Conclusion The results demonstratde that mice oocyte could be reconstructed by transferring cumulus cell into enucleated MⅡ oocyte and fertilized at the same time.The developmental potential of reconstructed embryo was poor as they blocked at 2 cell stage
