摘  要:为研究甲状旁腺素 (PTH)对大鼠成骨肉瘤细胞UMR10 6细胞增殖及 β 微管蛋白表达的影响。采用流式细胞技术测定DNA合成的变化并进行细胞周期分析 ,用Northernblotting方法观察β 微管蛋白的表达量。运用间接免疫荧光技术及Westernblotting方法观察微管聚合状态及微管蛋白的表达。结果显示 ,用PTH处理UMR10 6细胞 12小时后 ,S期细胞比例明显增大 ,提示PTH具有促该细胞株增殖作用。同时β 微管蛋白的mRNA及蛋白质表达量增加 ,β 微管聚合加强。表明 β 微管蛋白的合成及聚合与细胞增殖间可能存在着一定关系。In order to study the effect of parathyroid hormone (PTH) on rat UMR106 cell proliferation and β-tubulin expression, the change of DNA synthesis was determined and the cell cycle was analyzed by flow cytometry; the β-tubulin expression was observed by Northern blotting method; the microtubule polymerization and the tubulin expression were observed by indirect immunofluorenscent technique and Western blotting method. The results showed that 12 hours after the treatment of UMR106 cells by PTH, the percentage of S-staged cells was remarkably increased, suggesting that PTH functions to promote the proliferation of this cell strain, to increase the mRNA and protein expression of β-tubulin and to strengthen β-tubulin polymerization. It was indicated that there seems to be a certain relationship between the synthesis and polymerization of β-tubulin and the proliferation of the cells.
