摘 要:目的 评估复合骨髓间充质干细胞 (MSCs)的生物陶瓷在修复兔松质骨缺损中的作用。方法 分别对单纯煅烧骨、复合MSCs煅烧骨、三磷酸钙 (β TCP)及复合MSCsβ TCP行 :①体外研究 :电镜观察复合后MSCs的生物学行为 ;②裸鼠异位成骨研究 ;③兔股骨髁松质骨缺损 ( 6mm、深 12mm)修复 :18只兔 32侧骨缺损分为 5组 (其中一组未加任何处理 ) ,于 2、4、8周末分别行组织学检查。结果 ①MSCs在煅烧骨及 β TCP上的黏附及生长情况良好 ,4d基本长满表面 ,6d起开始重叠生长 ;②在裸鼠体内 ,单纯 β TCP或煅烧骨无成骨 ,而复合MSCs组自 2周开始成骨 ,并随时间延长而增加 ,但 β TCP组成骨与材料降解相平衡 ,而煅烧骨组仅在孔壁周围形成板层骨 ;③兔股骨髁缺损 :未经处理不能自行愈合 ,2周内各组均无骨长入 ,4周开始由周边长入 ,8周时各组的骨长入率分别为4 0 %、5 6 %、4 5 %、6 0 % ,无论在煅烧骨抑或 β TCP组中复合MSCs组的成骨量均高于对照组 (P <0 0 5 )。Objective To evaluate the role of calcium phosphate ceramics hybrided with mesenchymal stem cells(MSCs) in repairing cancellous bone defects in rabbits. Methods 4 kinds of implants including calcined bone, calcined bone hybrided with MSCs, β TCP (tricalcium phosphate) and β TCP hybrided with MSCs were studied respectively ① In vitro study: the biological activity of MSCs adhesion on the ceramics was observed with electronic microscopy; ②Tests in nude mice: bone formation after implantation was studied; ③Study on cancellous bone defect reparation in rabbits: 32 bone defects ( 6 mm, 12 mm in length) of 18 rabbits were divided into five groups, including one group without implantation. Histologic examination were performed at the end of 2nd?4th and 8th week. Results ① MSCs adhesion, growth and proliferation on the surface of calcined bone and β TCP were good. On the 4th day, confluence was formed, after 6 days MSCs formed multiple layers. ② Tests in nude mice: There was no osteogenesis in simple calcined bone or β TCP, while bone began to form at the 2nd week in the groups of ceramics hybrided with MSCs. However, the equilibrium of the osteogenesis and biodegradation was found in β TCP, while bone formation was limited to the wall of pore in calcined bone; ③Bone defect in rabbit femoral condyle: The defects without implantation couldnt heal spontaneously. There was no bone formation within 2 weeks, by the 4th week, osteogenesis began from the peripheral area. At the end of 8th week, the bone penetration ratio in the four groups were 40%, 56%, 45% and 60% respectively. The osteogenesis of ceramics hybrided with MSCs in both calcined bone and β TCP groups were significantly more than that of control group ( P <0 05). Conclusion Calcium phosphate ceramics hybrided with MSCs is an effective method for repairing cancellous bone defects.
