摘 要:为研究猪γ-干扰素在抗御传染性疾病和肿瘤生物治疗中的免疫增强作用,本实验以ConA刺激成华猪外周血淋巴细胞提取激活淋细胞的mRNA,设计合成特异的引物序列,应用反转录-聚合酶链式反应(RT-PCR)扩增和克隆成华猪IFN-γ基因并进行序列测定。成华猪IFN-γcDNA约长500bp,从第68位核苷酸开始编码其信号肽,到569位核苷酸为终止子。IFN-γ基因的开放框架(ORF)由498bp构成,编码166个氨基酸。成华猪IFN-γ与猪IFN-γ基因核苷酸间同源性为98%,在261-269存在点突变,在氨基酸水平完全一致。IFN-γ氨基酸与鼠同源性为30%,与人同源性为40%。In order to study the immunoadjuvant activity of IFN-γ gene of porcine, ConA was used to stimulate the peripheral blood lymphocytes for 24h. After the total RNA was isolated from the lymphocytes, the porcine IFN-γgene was amplified by reverse transcription polymerase chain reaction (RT-PCR). From the sequence, it was found that the gene of Chenghua porcine IFN-γwas consisted of 498 nu-cleotides,encoding 166 amino acids. The homology of IFN-γgene was 98% at nucleotide level between Chenghua porcine and other porcine. Porcine IFN-γgene has 40% homology with human IFN-γ, and less than 30% with mouse at amino acids level. The results showed that Chenghua porcine IFN-γ gene could be used as immunoadjuvant to raise the immunity of animals in further research work.
