盐酸氮斯汀对药物诱发豚鼠哮喘的影响

摘  要:目的 :观察盐酸氮 艹卓 斯汀 (AzelastineHydrochloride ,AZ)对豚鼠实验性哮喘的作用、药效学及药动学特点及作用机制。方法 :采用组胺和乙酰胆碱诱发在体豚鼠实验性哮喘模型。结果 :盐酸氮 艹卓 斯汀能够剂量依赖性地拮抗组胺和乙酰胆碱的引喘作用 ,明显延长引喘潜伏期 ,显著减少抽搐动物发生率 ,其ED50 为 0 .2 1 6mg/kg ,95 %可信限为 0 .2 1 4~ 0 .2 1 9mg/kg。结论 :预先给予盐酸氮 艹卓 斯汀能显著拮抗组胺和乙酰胆碱所诱发的在体豚鼠实验性哮喘。Objective To observe the effect of azelastine hydrochloride on experimental asthma and its pharmacodynamic and pharmacokinetic characteristics, as well as its mechanism. Methods Experimental asthma model on guinea pigs was induced by histamine and acetylcholine in vivo. Results Azelastine hydrochloride significantly inhabited asthma induced by histamine and acetylcholine in guinea pigs in a dose-dependent manner, obviously prolonged the incubation period of histamine and acetylcholine induced asthma( P <0.05), decreased the rate of tic of guinea pigs. The means effective dose(ED 50 ) is 0.216 mg/kg and its 95% of confidence limit is from 0.214 mg/kg to 0.219 mg/kg. Conclusion Azelastine hydrochloride could significantly inhabit the asthma induced by histamine and acetylcholine if it was used before the asthma.
