摘　　要：目的　体外扩增日本血吸虫中国大陆株黏蛋白样蛋白 (SjMLP)的部分基因序列。方法　利用PCGENE软件查找SjMLP的抗原决定簇 ;特定寡核苷酸引物的设计与合成 ;TRIZOL抽提日本血吸虫成虫总RNA ,RT -PCR扩增目的基因并进行DNA序列分析。结果　RT -PCR特异性扩增出SjMLP编码区部分基因序列 ,大小为 75 6bp ,测序结果显示与曼氏血吸虫MLP高度同源。结论　扩增到了SjMLP抗原编码区基因的部分序列 。Aim To provide the basis for further study of DNA vaccine against schistosoma japonicum.Methods The antigenic determinants were predicted by using PCGENE software and specific oligonucleotide primers were designed and synthesized;Total RNA was isolated from adult worms of Chinese S.japonicum by using TRIZOL reagent;Coding region of SjMLP gene was amplified by RT-PCR;Results The coding region of SjMLP gene was specifically amplified by RT-PCR and the size of amplified fragment was 756 base pairs. The DNA sequencing resuslt indicated that the part coding sequence of SjMLP is highly homology to the sequence of schistosoma mansoni.Conclusion Sequence of SjMLP is highly homology to of schistosoma mansoni.
