摘  要:目的阐明有氧运动抑制或延缓动脉粥样硬化(atherosclerosis,AS)具体作用机制,探讨有氧运动在AS形成过程中的作用。方法将30只C57B2/6J纯系小黑姒按序号分为政党对照组,AS组和高脂膳食+运动组(各10只,)应用免疫组织化学SABC法并结合血脂水平及超微结构改变,对实验性高胆固醇血症小鼠主动脉壁血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)蛋白表达及有氧运动对其表达的影响进行观察。结果在致AS的高胆固醇饮食条件下,AS组小鼠主动脉壁的血栓凝块内及增生内膜的尖端或侧壁均有VEGF强表达。而在同样条件下经8周有氧耐力训练的HE组小鼠主动脉壁却无血栓及内膜增生,仅局部内皮细胞出现VEGF强表达。电镜显示AS组小鼠内皮细胞体积增大,内有空泡。平滑肌细胞内粗面内质网及高尔基体增多,弹力纤维变形。而HE组接近正常。HE组的三酰甘油(TG),总胆固醇(TC),低密度脂蛋白(LDL)均低于AS组(t=2.132,2.215,2.943,P<0.01);高密度脂蛋白(HDL)高于AS组(t=2.196,P<0.01)。结论内源性VEGF与AS的发生发展有关;有氧运动除可降低血脂外,同时降低血管壁VEGF表达而抑制AS形成。Aim To discuss the effect and mechanism of aerobic exercise in inhibiting the formation of atherosclerosis(AS). Methods Thirty C57BL/6J black mice were randomly divided into three groups of 10 subjects in sequence: normal control group (NC group), atherosclerosis group (AS group), aerobic exercise group (AE group). NC group was fed with normal forage, while AS group and AE group were fed with normal forage plus high fat diet. Blood lipids were measured, including total cholesterol (TC), triglycerides(TG), high density lipoprotein(HDL), low density lipoprotein (LDL); vascular endothelial growth factor (VEGF) protein expression in aorta wall was measured with immunohistochemical SABC method; the microstructures of ascending aorta were objected though TEM. Results The expression of VEGF protein in AS group was intensive in the fibrin clot of thrombus and the top or side of the proliferating neointima in aorta wall, but that in AE group with 8weeks aerobic exercise was only intensive in local endothelial cells. The endothelial cell became bigger with vacuoles, and rough endoplasmic reticulum (RER) and Golgi apparatus increased with the deformation of elastic fibers in comparison with those in AE group. TG, Tc and LDL in AE group were lower than those in AS group(t=2.132,2.215,2.943,P< 0.01£(c), and HDL in AE group was higher than that in AS group(P< 0.01). Conclusion The entrogenous VEGF is related to the formation of atherosclerosis. Besides decreasing plasma lipid, aerobic exercise might decrease the expression of VEGF in blood vessel wall so as to inhibit the formation of atherosclerosis.
