老年高血压合并2型糖尿病患者血小板表面P-选择素及糖蛋白Ⅱb/Ⅲa的变化

摘　　要：探讨老年高血压合并 2型糖尿病患者血小板膜上P 选择素 (CD6 2 )及糖蛋白Ⅱb/Ⅲa(PAC 1 )的变化 ,采用流式细胞术 (FCM)以单克隆抗体分子作为探针检测健康老年人及高血压合并 2型糖尿病患者血小板膜上CD6 2的阳性百分率 (CD6 2 % )、CD6 2的几何荧光强度 (CD6 2MFI)、PAC 1的阳性百分率及PAC 1的几何荧光强度的表达量。结果显示 ,老年高血压合并 2型糖尿病患者血小板膜上CD6 2 %、CD6 2MFI、PAC 1 %及PAC 1MFI的表达量明显升高 ,但与当日血压、血糖及血脂无相关关系。研究表明 ,CD6 2及PAC 1是血小板活化的敏感指标 ,高血压合并 2型糖尿病患者有血小板活化现象 ,为高血压合并 2型糖尿病患者除了充分降压、降脂及降糖治疗外 。To investigate the changes in P selectin( CD62) and glycoprotein Ⅱb/Ⅲa (PAC 1) on platelets in elderly patients with hypertension and type 2 diabetes. CD62 and PAC 1 on platelets were analyzed by flow cytometric 3 color immunofluorescence. The percentage and MFI of CD62 expression on the platelet membrane in elderly patients with hypertension and type 2 diabetes was increased compared with healthy individuals(6.36 vs 0.91 P <0.01,43.68 vs 26.70 P <0.05). The percentage of PAC 1 and MFI expressed on the platelet membrane were also increased (23.60 vs 1.90, P <0.01,45.6 vs 23.50, P <0.01). All of them were not related to blood pressure or the contents of blood lipid and glucose. These results suggested that CD62 and PAC 1, especially PAC 1,could be regarded as sensitive and specific molecular markers for activated platelet. The antiplatelet drug is needed in elderly patients with hypertension and type 2 diabetes besides antihypertensive, antilipemic and antidiabetic drugs.
