摘 要:目的 :研究不同浓度的过氧化氢对培养的人腹膜间皮细胞 (humanperitonealmesothelialcells,HPMC)产生趋化蛋白 -百介素 - 8(IL - 8)、单核细胞趋化蛋白 - 1(MCP - 1)的影响以及高糖环境下、抗氧化剂丙酮酸存在时过氧化氢作用的改变。方法 :分别在不同浓度的葡萄糖环境下 ,用不同浓度外源性小剂量过氧化氢、丙酮酸刺激细胞 ,用半定量PT -PCR方法测定IL - 8、MCP - 1基因水平 ,用ELISA方法测定IL - 8表达。结果 :(1)亚毒剂量的过氧化氢可以使HPMC表达IL - 8增加 ,但对MCP - 1mRNA的影响不大。且高糖环境下 ,外源性过氧化氢能够使HPMC产生更多的IL - 8。 (2 )加入丙酮酸后 ,IL - 8的表达下降 ,而MCP - 1的表达未见明显的改变。结论 :(1)外源性小剂量的过氧化氢可使HPMC产生IL - 8增加 ,且高糖环境使之作用增强 ,可能增加透析液的生物不相容性。 (2 )丙酮酸能够对抗氧化应激 。Objective:To study the effects of different concentration hydrogen peroxide on the production of IL-8 and MCP-1 in human peritoneal mesothelial cells(HPMC)in vitro. Moreover to research the influence of adding high concentration glucose and pyruvate in the roles of hydrogen peroxide.Methods: The glucose(0.1%? 3.86%)?the hydrogen peroxide(0.1 mmol/L?0.5 mmol/L) and pyruvate (35 mmol/L) were used in culture medium to stimulate the HPMC.IL-8(Interlukin-8)mRNA and MCP-1(Monocyte chemoattractant protein-1)mRNA were measured by semiquantitative RT-PCR,moreover,the protein level of IL-8 by ELISA.Results:(1)Small dose hydrogen peroxide increased the expression of IL-8,both of mRNA and protein,which was lowered by the antioxidant and increased by the high glucose.But both the small dose of hydrogen peroxide and the antioxidant had no influence in the expression of MCP-1 mRNA.Conclusion:Exogenous sub-toxic dose hydrogen increase the expression of IL-8 and high glucose enhance this effect,which may augment the uncompatibility of peritoneal fluid,adding the antioxidant(pyruvate)to the patient of CAPD may be useful.
