野败型水稻保持系线粒体特异DNA片段的克隆及序列分析

摘　　要：利用RAPD技术 ,从水稻野败型细胞质雄性不育系珍汕 97A的保持系珍汕 97B中得到一个特异的扩增片段PWP 13。该片段全长 80 8bp ,1～ 14 2区段为正常的cob基因片段 ;14 3～ 372、4 0 6～ 70 7区段与一个报告的嵌合cob基因的同源性分别为 98%、10 0 % ,但由于碱基缺失 ,所推测的氨基酸序列差异较大 ;373～ 4 0 5区段为PWP 13所特有的重复序列 ,70 8～ 80 8区段为未知序列。序列内含有 3组长度分别为 9、31、2 7bp的小重复序列。A mtDNA fragment specific to Zhenshan 97B, the maintainer line of a wild abortive type male sterile line, Zhenshan 97A, was amplified by RAPD. It is 808 bp in length(Figs.1-3). The nucleotide sequences located at the positions from 1 to 142 are identical with some parts of sequences in a apocytochrome b. The nucleotide sequences from 143 to 372 and from 406 to 707 have high homology with a pseudo apocytochrome b and identity rates reach to 98%, 100% respectively, but the derived amino acid sequence is different due to the loss of some bases. The nucleotide sequences from 708 to 808 are unknown(Figs.4,5,Table 1). The nucleotide sequence of PWP 13 contains 3 small repeated sequences 9, 31 and 27 bp in length(Fig.4).
