沥水调脂胶囊对弱氧化型低密度脂蛋白诱导血管平滑肌细胞c-jun、c-fos、c-myc mRNA表达的影响

摘　　要：目的 :探讨沥水调脂胶囊抑制血管平滑肌细胞增殖的机制。方法 :用消化法获取血管平滑肌细胞 (VSMC) ,DMEM培养 ,以甲醇除蛋白后的沥水调脂胶囊含药血清、弱氧化型低密度脂蛋白(mox LDL)和vitE分组处理细胞 ,RT PCR扩增c jun、c fos、c myc ,用凝胶成像分析仪分析琼脂糖电泳结果 ,与GAPDH对照进行半定量分析。结果 :在处理后 30min含药除蛋白血清和vitE对mox LDL诱导VSMCc fos、c jun、c mycmRNA表达均无明显影响 ,在 1h、2hvitE组与对照组比较均有明显差异 (P <0 0 5 )。含药除蛋白血清组c myc、c fos在 1h、2h与对照组比较无明显差异 ,c jun则明显降低 (P <0 .0 5 ) ,与vitE的作用不相同。结论 :降低JNK/SAPK途径信号途径可能是沥水调脂胶囊抑制VSMC增殖的机制之一。Objective: to study the pharmacology of Li Shui Tiao Zhi capsule inhibited Vascular Smooth Muscle Cells induced by Mildly Oxidized Low Density Lipoprotein in vitro. Method:serum containing medicine removed protein,vitamin E and mox LDL treated vascular smooth muscle cells;reverse transcription polymerase chain reaction analysis of c fos,c jun,c myc mRNA levels. Results: Vitamin E significantly reduced the moxLDL induced expression of c fos,c jun,c myc mRNA at 1 and 2h; the levels of c fos and c myc mRNA were not significantly affected by serum containing medicine removed protein,but serum containing medicine removed protein significantly reduced the moxLDL induced expression of c fos,c jun,c myc mRNA at 1 and 2h. Conclusion: Effects on upper streams of JNK/SAPK signal transduction pathway may partly contribute to the antiproliferation of Li Shui Tiao Zhi capsule in atherosclerosis.
