摘 要:目的:探讨抗纤复方含药血清对肝星形细胞(HSC)L190细胞(HSC-LI90)Ⅰ型、Ⅳ型前胶原和基质金属蛋白酶基因表达的影响。方法:以0.5、2.0、4.0g/kg等不同剂量抗纤复方灌胃,制备大鼠的含药血清,作用HSG-LI90细胞48小时,应用Northem印迹杂交的方法,检测Ⅰ型、Ⅳ型前胶原、基质金属蛋白酶-2及膜型基质金属蛋白酶基因的表达,并用酶图法检测基质金属蛋白酶-2的活性。结果:抗纤复方含药血清能抑制HSC-LI90细胞Ⅰ型、Ⅳ型前胶原、增加模型基质金属蛋白酶基因表达,对基质金属蛋白酶-2基因表达及活性无影响。结论:抗纤复方能下调Ⅰ型、Ⅳ型胶原的合成,增加模型基质金属蛋白酶基因的表达。Objective: To study the effects of anti-fibrosis compound (AFC) contained serum on type I and Ⅳ procolla-gen and matrix metalloproteinase (MMP) gene expression in HSC-LI90 cells. Methods: HSC-LI90 cells were exposed in different doses of AFC contained serum (0.5, 2.0, 4.0g/kg) for 48 hours. The gene expressions of type I and Ⅳ procollagen and matrix metalloproteinases-2 (MMP-2) and membrane type matrix metalloproteinase (MT-MMP) were investigated by Northern blot. The gelatinase activity of MMP-2 was examined by zymography. Results: AFC contained serum could inhibit the type I and Ⅳ procollagen and increased the MT-MMP the gene expressions of HSC-LI90. The expressions of MMP-2 and MMP-2 activity were not affected by different doses AFC contained serum. Conclusion: The results demonstrate that AFC contained serum can down-regulate type I and IV collagen synthesis and increased the MT-MMP gene expression.
