针刺对面神经再生逆行轴突转运神经营养因子的影响(英文)

摘　　要：目的通过观察针刺对面神经逆行轴突转运神经营养因子的影响,以从分子生物学水平探讨针刺促进周围神经再生的机制。方法同龄成年健康的新西兰兔180只,雌雄不限,体质量1.8～2.8kg。均在30mg/L的戊巴比妥钠静脉麻醉下,分离暴露面神经上颊支,在手术放大镜下切断神经,用硅胶管将两断端嵌入并缝合固定,形成再生室。术后动物根据体重和雌雄,按随机数字表分为针刺组和对照组,每组各90只,每组的90只又以随机数字表法分为3小组,每小组各30只。分别接受125I-BDNF,125I-NT-3或125I-NT-4再生室内注射,10μl/只。针刺组于术后2和26h各接受1次治疗,对照组不作任何处理。术后4,8,24,48h摘取含面神经核的脑干检测,两组对照。结果对照组脑干中125I-BDNF,125I-NT-3或125I-NT-4的γ计数在12和24h达到峰值,分别为犤(1413±1),(1348±4)〗,犤(1352±3),(1286±3)〗犤(806±2),(1137±2)〗,48h又明显降低,分别为(594±1),(624±2),(354±2),不同时段波动很大。而针刺组脑干的125I-BDNF,125I-NT-3,125I-NT-4γ计数在各个时段始终保持在较高的水平。结论针刺可促进面神经神经营养因子逆行轴突转运。Aim To explore the mechanism of acupuncture promoting regeneration of peripheral nerve by observing the effect of acupuncture on facial nerve regeneration neurotro phic factor.Methods 180Newzeland rabbits with the same age and weighed1.8-2.8kg were selected and un-derwent venous anesthesia by 30mg /L sodium pentobarbital.Upper bucal branch of facial nerve was separated,exposed and cut under operating mag-nifying glass.The ends were inlaid i nto silica gel tube to form regeneration room.After operation,rats were div ided by randomly according to body ma ss and sex into acupuncture group and co ntrol group with 90rats in each group.In each group,90rabbits were also divided into 3subgroups random ly with 30rabbits in each and underwent 125 I-BDNF,125 I-NT-3or 125 I-NT-4re-generation intraventrical injection,10μl /rabbit.Teh rabbits in acupunctur e group received treatments at 2and 26h respectively while rabbits in cont rol received no treatment.At 4,8,24,48h,brain stems containing nucleus of facial nerve were taken and examined.Resultsγcounts of 125 I-BDNF,125 I-NT-3or 125 I-NT-4in control group got to climax at 12,24and were(1413±1),(1348±4),1352±3),(1286±3)(806±2),(1137±2)]respectively,but decreasedat48 hand were(594±1),(624±2),(354±2).Fluctuation was obvious.While in acupuncture group,γcounts of 125I-BDNF,125 I-NT-3 or 125 I-NT-4 were kepta higher level at each timepoint.Conclusion Acupuncture can promote facial nerve retrograde ax-is-cylinder transport nurotrophic factor
