摘  要:目的 比较从人外周血单核细胞及脐血 CD34 +细胞诱导树突细胞 (DC)的方法及其特性。 方法 用淋巴细胞分离液分离外周血单个核细胞 ,贴壁法获得单核细胞 ;经 GM- CSF及 IL- 4诱导获得 DC。磁性活化细胞分选系统 (MACS)分选脐血 CD34 + 细胞 ,以 GM- CSF、IL - 4、TNF-α、SCF、FL、TPO等细胞因子诱导获得 DC。用电镜、普通显微镜观察 DC形态 ;流式细胞仪分析 DC表型 ;同种混合淋巴细胞反应 (allo- ML R)观察 DC刺激 T淋巴细胞增殖的能力。 结果 外周血单核细胞在因子 GM- CSF、IL - 4、TNF-α,脐血 CD34 + 细胞在 GM- CSF、IL - 4、TNF-α、SCF、FL、TPO等因子作用下 ,可生成 DC,并表达相应的 DC分化抗原 CD1 a、CD80 、CD83、CD86 、HL A- DR。脐血 CD34 + 细胞具有较强的扩增能力 ,经 2周诱导 ,体系细胞数可增加 173.8± 2 6 .7倍 ,两种来源的 DC均能刺激同种异体淋巴细胞的增殖反应。 结论 外周血单核细胞与脐血 CD34 +细胞 ,在体外均可诱导成 DC,并具有相应的Objective To compare the induced dendritic cells(DC) from human peripheral blood monocytes(PBMC) and umbilical cord blood(CB) CD\-\{34\}\++ cells, and investigate it's ability to stimulate T cell proliferation. Methods Mononuclear cells were isolated from peripheral blood with Ficoll solution and monocytes were obtained by adherent treatment; CD\-\{34\}\++ cells were isolated from umbilical cord blood by using magnetic activated cell sorting(MACS) system. Dendritic cells from PBMC were induced with GM\|CSF, IL\|4, and those from CB were induced with GM\|CSF, IL\|4, TNF\|α, SCF, FL, TPO. The morphology of DC was observed under the electron microscope and optical microscope. The phenotype of DC was analyzed by flow cytometer. Proliferation ability of T lymphocytes was tested by allogenic mixed lymphocyte reaction(allo\|MLR). Results DC can be induced from PBMC with GM\|CSF, IL\|4, TNF\|α, and from CB\|CD\-\{34\}\++ cells with GM\|CSF, IL\|4, TNF\|α, SCF, FL, TPO. Both of DCs expressed CD\-\{1a\}, CD\-\{80\}, CD\-\{83\}, CD\-\{86\}, HLA\|DR. DC from CB CD\-\{34\}\++ cells had strong expansion ability in vitro with 173.8±26.7 times after two weeks culture. Both of DCs which were induced from two kinds of different sources can stimulate allogeneic lymphocytes proliferation reaction. Conclusion DC could be induced from peripheral blood monocytes and cord blood CD\-\{34\}\++ cells, and possessed the abilities to stimulate T lymphocytes proliferation.
