自制抗原片检测抗角质蛋白抗体的结果分析及价值

摘　　要：目的　探讨自制抗原片检测抗角质蛋白抗体 (AKA)价值。方法　分别以自制Waster大鼠食管抗原片和德国欧蒙公司抗原片为底物 ,用间接免疫荧光法检测AKA ,并比较Even氏蓝复染与不采用Even氏蓝复染对结果的影响。结果　两种抗原片在阳性率和非特异性荧光出现率方面均无统计学差异 (P >0 .0 5 ) ,Even氏蓝复染与否也无统计学差异 (P >0 .0 5 ) ;类风湿性关节炎 (RA)组与非RA组AKA阳性率差异非常显著 ( P <0 .0 1) ;AKA对RA的诊断有较高的敏感性和特异性。结论　自制Waster大鼠食管抗原片 ,经济实用 ,适合临床应用 ;Even氏蓝复染与否对结果无影响 ;Objective To build up a detection method of antikeratin antibodies (AKA) for clinical application.Methods AKA was detected by indirect immunofluorescence (IIF) in two kinds of straum corneum of six week old Wistar rat esophagus:one was made by ourselves,another obtained from Germen Euroimmun Com.Results (1)There was no significant difference in positive rate of self made sections,as compared with Euroimmun slides (P> 0.05), and nor was in counterstaining (P>0.05).(2)Statistical difference was found between RA group and non RA group (P<0.01).The sensitivity and specificity of AKA were quite high.Conclusion (1)Self made frozen sections for detecting AKA should be used more widely by IIF technique.(2)There is no influence on the result whether Even's blue counterstaining or not.(3)AKA has a important value for early diagnosis.
