Fas及FasL表达对大鼠睾丸局部热作用生精细胞凋亡的影响

摘　　要：目的 :探讨 Fas及 Fas L表达对睾丸局部热作用后生精细胞凋亡的影响。方法 :在大鼠睾丸局部受热后 0 .5 d、1 d、3d和 6d,应用流式细胞仪、原位末端标记法 ( TUNEL)检测生精细胞凋亡情况 ;应用流式细胞仪和免疫组化法检测 Fas和 Fas L蛋白表达情况。结果 :与其对照组相比热处理各组生精细胞凋亡明显升高 ( P<0 .0 1 ) ,各热处理组 Fas表达明显增强 ( P<0 .0 1 ) ,Fas L表达亦增强 ( P<0 .0 5 ) ;并且热处理后 Fas及 Fas L阳性表达增强和凋亡增多在发生时间和生精细胞的定位上基本吻合。结论 :睾丸局部热作用可引起生精细胞凋亡增多 ;Fas和 FasObjective: To study Fas/FasL expression and spermatogenic cell apoptosis in rat with local testicular heating. Methods: The apoptosis of spermatogenic cells was detected by means of flow cytometry and TUNEL (TdT mediated dUDP X nick end labeling) method and Fas/FasL expression were observed with flow cytometry and immunohistochemistry on hours 12 and days 1, 3 and 6 after heat exposure. Results: In all the experimental groups, the apoptosis of spermatogenic cell was significantly increased( P <0.01)and Fas/FasL expression were highly increased( P <0.01, P <0.05 respectively) compared with the respective control group. Moreover, The enhancement of Fas/FasL expression approximately coincidce with the apoptosis increase in the time and the localization of spermatogenic cell after heat exposure. Conclusion: These findings suggest that Local testicular heating could enhance the spermatogenic cell apoptosis; Fas/FasL are importnt apoptosis regulating genes that play an important role in spematogenic cell apoptosis after heat exposure.
