摘  要: 目的:建立分泌抗海洋褐胞藻(Chattonellamarina)、卵圆褐胞藻(Chattonellaovata)、赤潮异弯藻(Heterosigmaakashiwo)、具齿原甲藻(Prorocentrumdentatum)、尖刺拟菱形藻(Pseudonitzschiapungens)等5种藻的高特异性单克隆抗体杂交瘤细胞株.方法:利用细胞融合技术制备杂交瘤细胞,并通过反相间接血凝、间接酶联免疫吸附测定(ELISA)等方法筛选克隆阳性杂交瘤细胞株.结果:获得15CA7、23BG2和23BG9等3株杂交瘤细胞分泌的单克隆抗体与5种藻具有一定的特异性,但抗体的亲和力较低,仅与海洋褐胞藻具有较高的亲和力;而获得的M18杂交瘤细胞株分泌的单克隆抗体可与以上除尖刺拟菱形藻外4种藻产生高效价的明显交叉反应.结论:获得了3株分泌对5种藻具有一定的特异性较低亲和力抗体;而M18细胞株分泌的单抗与除尖刺拟菱形藻外4种藻产生明显的高效价的交叉反应,显示此单抗可代替5种海洋藻的抗血清.Aim: To obtain several strains of cells which can secrete highly specific monoclonal antibodies against five kinds of algae which are Chattonella mar ina, Chattonella ovata, Hetero-sigma akashiwo, Prorocentrum dentatum and Pse ud onitzschia pungens, respectively. Methods: The hybridomas were obtained by cell fusion technique and monoclonal antibodies selected by indirect ELISA, reverse indirect agglutination. Results: The three strains of clones, 15 CA7, 23BG2 and 2 3BG9,which can secrete specific monoclonal antibodies against Chattonella mari na . And clone M18 can secrete monoclonal antibodies which can have significan t cross- reactivity with 5 kinds of algae showed above. Conclusion: It is feasib le to use the 3 hybridomas we have obtained to produce highly specific m onoclon al antibodies which can be used to identify and determine Chattonella marina . That monoclonal antibodies secreted by clone M18 showed high cross-reactivity am ong 5 kinds of algae indicates the monoclonal antibodies can be used as polyclon al antibodies against 5 kinds of algae. 
