大鼠实验性糖尿病免疫组织化学观察

摘　　要：本实验以四氧嘧啶50mg/kg尾静脉注射的方法制成大鼠实验性糖尿病动物模型。用HRP—SPA免疫组织化学方法观察了大鼠患实验性糖尿病后胰岛B细胞的动态变化。结果表明,注射四氧嘧啶后48h每个大鼠的几乎全部胰岛均发生明显的形态学改变,表现为胰岛形态欠规则。细胞极性紊乱,除少数胰岛尚可见数个散在的胰岛素免疫反映阳性细胞外,多数胰岛内胰岛素免疫反应阳性产物缺失与消失,多数细胞核固缩、变形。给药7天后的大鼠胰岛普遍出现散在的免疫反应阳性细胞,数目也较伤后48h时增多,并偶见免疫反应阳性细胞密集分布的胰岛。提示四氧嘧啶致实验性糖尿病一周后,胰岛B细胞有逐渐恢复或再生的迹象。In this study, the animial models with experimental diabetes were prepared by iv. alloxan (50mg/kg). At the 48th hour and on the 7th day after injection of alloxan, We separately observed the island B cell changes using HRP -- -- SPA immunohistochemistry stain. The results suggested that at the 48th hour after injection of alloxan, all island were almost injured, with irregular appearance, derangement polar cells, the deficiency and disappearan ce of immunoreactive insvlin positive products, pyknosis and deformation in most nucleus, at the same time only in a few islands exist several immunoreactive insulin positive cells and that on the 7th day after injection of alloxan, most of the islands appeared scattered immunoreactive positive cells, were seen incidentally. It was suggested that experimental diabetes induced by alloxan have the ability to recover gradaully.
