Morphological and morphometric study of rat spiral ganglion neurons during postnatal development

摘  要:目的 证实出生后第 2周是大鼠耳蜗螺旋神经节神经元发育的关键时刻。方法 选用 60只出生后 1~ 49d( postnatalday 1to 49,PND 1~ 49)的SD大鼠 (分为 10组 )和 6只成年 ( 4月龄 )SD大鼠。取平行蜗轴的中轴切片 ,光镜下观察螺旋神经节神经元形态 ;通过形态学测量神经元细胞面积、细胞核面积、Rosenthal隧道截面积并计数神经元 ,计算核浆比和细胞密度。结果 PND 1~ 14大鼠神经元细胞面积逐渐增大 ;PND 1~ 3大鼠细胞核面积较小 ,此后无明显变化趋势 ;PND 7大鼠细胞密度和核浆比明显下降 ,PND 14大鼠核浆比有明显下降。发育最早从钩回开始 ,逐渐向顶回发展。结论 出生后第Objective To confirm that the second postnatal week of the rats is the critical period for the development of the cochlear spiral ganglion neurons. Methods 60 Sprague Dawley rat pups aged between postnatal day 1 and 49 (divided into 10 groups) and 6 adult rats were used in the study. The parallel mid axes sections were used to observe the morphology of spiral ganglion neurons under light microscope. Morphometric data were obtained with a high resolution semiautomatic image analysis system. The following parameters were measured: ① neuronal cell and nuclear area, ② nuclear to cytoplasmic ratio, and ③ Rosenthal's canal area and number of neurons per surface unit. Results The area of the cell became larger and larger from PND 1 to PND 14;The nuclear area in PND 1~3 was small, from then on with presence of no evident changes. The cell density and the nuclear to cytoplasmic ratio in PND 7 declined significantly, and in PND 14 the nuclear to cytoplasmic ratio declined further more. The maturation of the neurons sequentially progressed from the extreme basal (hook), then gradually to the apexturn of the cochlea. Conclusion The second postnatal week of the rats is the critical period for the development of the cochlear spiral ganglion neurons.
