摘 要:目的 建立酸水解 酸性染料比色法同时测定贝母类药材中生物碱及生物碱苷含量的方法。方法 用氯仿 甲醇 (4∶1 )混合溶剂提取总碱 ,酸性染料比色法测定水解前及水解后提取物中生物碱含量 ,计算出生物碱、生物碱苷及总生物碱的含量。结果 生物碱、生物碱苷及总生物碱的加样回收率分别为 1 0 0 .5 % ,1 0 2 .2 % ,1 0 1 .1 % ,RSD分别为 1 .1 % ,3 .3 % ,1 .7% (n =5) ,并对药典收载的 9种贝母类药材进行了测定。结论 本法不需要复杂仪器 。OBJECTIVE: To develop the acid hydrolysis-acidic dye color-metric method for the determination of alkaloid and glucoalkaloid in Bulbus Fritillariea. METHODS: The total alkaloid in Bulbus Fritillariae was extracted by chloroform-methanol (4:1). The contents of alkaloid, gluco-alkaloid and total alkaloid were calculated through the determination of alkaloid concentrations in extracts which was hydrolyzed or without hydrolyzed. RESULTS: The recovery of spiked alkaloid, gluco-alkaloid and total alkaloid were 100.5%, 102.2% and 101.1% respectively. The RSD were 1.1%, 3.3% and 1.7% (n = 5) respectively. Nine bulbus Fritillariae which were included in China Pharmacopeia were determined with this method. CONCLUSION: The method is simple without complex equipment. It could be used for the determination of alkaloid and gluco-alkaloid in Bulbus fritillariae.
