家蚕浓核病的血清学诊断方法

摘　　要：本文介绍了家蚕浓核病血清学诊断的系列方法,包括双向免疫扩散法(DID),对流免疫电泳法(CIEP),酶联对流免疫电泳法(ELCIEP),酶标组化法(IEHC),可溶性酶——抗酶法(PAP法),点免疫结合测定法(DIBA),生物素——亲和素系统(ABS)及胶乳凝集试验法(PAT)等,应用这些方法可以检测出的纯化浓核病毒(DNY)量(即检测灵敏度)为1×10^(-1)-1×10^(-4)O.D,以DIBA法检测灵敏度最高,分别为LAT法,ELCIEP法,CIEP法的10倍,160倍、1000倍,检测蚕体内的DNV时,可以在经口接种DNY后8-40小时检出阴性反应,以ABS法检出阳性反应最早,而在同样接种病毒的条件下,要到感染后5-6天才表现出明显的外观症状,试验表明,检出阳性反应时间越早,检出阳性率愈高,则最终发病率高,因此这些诊断方法可作为诊断家蚕浓核病的有效手段。This paper introduces a scries of methods of scrological diagnoses for the dcnsonuclcosis disease of the silkworm. They include double immunodiffusion (DID), counter immunoclcctrophorcsis (CIEP), enzyme-linked counter immunoclcxtrophorcsis (ELCIEP), immunocnzymic histochcmical method (TEHC), pcroxidasc anti-pcroxidasc method (PAP), dot immunobinding assay (DIBA), avidin- biotin system (ABS) and the latex agglutination test (LAT), etc. The amount of the purified dcnsonuclcosis virus (DNV) detected by these methods, i. e. the sensitivities of the detections, is 1×10^(-1)- 1×10^(-4)O.D. DIBA is the most sensitive method and its sensitivity is 10,160 and 1,000 times higher than that of LAT, ELCIEP and CIEP respectively. When these methods arc used to detect DNV in the silkworm, the positive reactions can be detected in 8-40 hours after the DNV- inoculation per os. The positive reaction by ABS appcrars earlier than by other methods. But the exterior symptoms of the diseased silkworms do not appear until the fifth or sixth day after the DNV- inoculation under the same situation. The tests have shown that the earlier the positive rcacgions appcrar and the higher the positivevc rale is, the higher the final incidence of the diseased silkworms is. Therefore, these methods can be used as an effective means for diagnosing the dcnsonuclcosis of the silkworm.
