摘 要:对11个大白菜地方种质材料,用改进的CTAB法提取总DNA后进行RAPD(RandomAmplifiedPolymorphicDNA)分析。聚类结果表明,当遗传距离为0 18时,11份材料可分为二个生态类群,第一类群属于直筒类型(交叉性气候生态型),另一类群在0 15时,又分为二个类群,矮桩类型(海洋性气候生态型)和直筒与矮桩的过渡类型。同时表明,应用RAPD选择特定引物在同一亚种内不同品种间进行遗传多样性、亲缘关系及品种鉴定等研究是可行的。RAPD analysis was used to evaluate polymorphism and genetic relationship of 11 local germplasm of Chinese cabbage.The cluster analysis results showed 11 materials were divided into two ecological groups when genetic distance was 0.18,one was Crossed Climate Ecotype and the another was subdivided into two subgroups when distance was 0.15,one was Ocean Climate Ecotype and the another was transitional form.Meanwhile,it was possible to study genetic relationship and polymorphism by RAPD with specific primer within subspecies.
