摘  要:〔目的〕观察以线粒体膜电位 (MitochondrialMembranePotential,MMP)标记法检测新风胶囊对佐剂性关节炎 (ad juvantarthritis,AA)大鼠滑膜及胸腺细胞凋亡的影响 ,探讨新风胶囊的作用机制。〔方法〕采用弗氏完全佐剂造成佐剂性关节炎 (AA)大鼠模型 ,设立正常对照组、模型对照组、甲氨喋呤 (MTX)对照组、雷公藤多甙 (TriperygiumWilfordiiPolycorideTablet ,TPT)对照组和新风胶囊 (XinfengCapsule,XFC)治疗组。记录各组大鼠的关节炎指数 ,应用流式细胞仪 (FCM)以线粒体膜电位 (MMP)标记法测定各组大鼠滑膜及胸腺细胞凋亡率。〔结果〕与治疗前相比 ,新风胶囊组、雷公藤组及MTX组治疗后大鼠的关节炎指数显著降低 (P <0 .0 5~ 0 .0 1) ;模型组滑膜及胸腺细胞凋亡率显著低于正常对照组 (P <0 .0 5 ) ,新风胶囊组细胞凋亡率显著高于模型组、雷公藤组及MTX组 (P <0 .0 5~ 0 .0 1)。〔结论〕新风胶囊与MTX、雷公藤一样能降低AA大鼠关节炎指数 ,在促进线粒体电位介导的滑膜及胸腺细胞凋亡方面优于MTX和雷公藤。促进滑膜细胞凋亡 ,抑制滑膜增生以及促进胸腺细胞凋亡 ,抑制自身免疫反应可能是新风胶囊降低关节炎指数、清除关节炎肿胀的机理之一。睴bjective〕 To observe the effect of Xinfeng capsule on the apoptosis of the Synoviocytc and thymus_lymphocyte of adjuvant arthritis(AA)rat by the Mitochondrial Membrane Potential(MMP).〔mcthods〕Model rats were established by the Freund's complete adjuvant.The rats were divided into 5 groups:normal control、model control、methotrexate(MTX)control、triperygium wilfordii polycoride tablet(TPT)control、cifeng capsule treated(XFC),The arthritis index(Al)was reeorded.The apoptptic percentagc of the synoviocyte and thymus_lymphocyte of AA was measured by floe cytometry(FCM)using MMP.〔Rcsults〕 Xinfeng capsule、MTX and TPT can decrease the AI( P <0.01~0.05).Compared with the normal control,apoptotic pcrcentage of the synoviocytc and thymus_lymphpcytc of model control was dccrcascd( P <0.05);Compared with the MTX、TPT and model control,apoptotie percentage of XFC was significant increased ( P <0.05~0.01).〔Conclusion〕Xinfeng capsule can decrease the AI like MTX、TPT.But it is superior to MTX、TPT in increasing the apoptosis of the synoyiocyth and thymus_lymphocte of AA.It maybe the mecbanism for XFC to dccrcasc AI that promot the apoptosis of synoviocyte ahd thymus_lymphocyte,restrain the synovial membrane from hypcrplasia and autoimmunc reaction.
